WebDec 12, 2024 · Tamoxifen is mainly a prodrug as two of its many metabolites, notably 4-hydroxy-N-desmethyltamoxifen (endoxifen) and also 4-hydroxytamoxifen (4-OHT), are known to have greater affinity for the estrogen receptor than tamoxifen itself. Routine therapeutic dosage starts at 20 mg daily and can be increased up to 40 mg daily. WebOf note, tamoxifen can also cause retinopathy.33 Retinal changes occur more frequently than corneal changes at high doses (180–320 mg/day),34 resulting in crystal-like deposits around the macula with macular oedema and granular changes in the retinal pigment epithelium, leading to impaired visual acuity.32 34
